How to remplace the main image by a video?

Yes. I would like to create 50 posts with 50 different videos (and display the video instead of the main image).

The simplest method would be to disable the Header Element on your posts and the featured image. And simply add a Video block to the top of your posts.

I tried, however the elements stay.

in Appearance > Elements you will need to edit the Single Post header element. And change its Display Rules to EXCLUDE that post. I would suggest using a Tag to identify the Video posts, then you can exclude the Post Tag

Is it possible to have a 100% width on a specific category ? e.g, here I would like that the content (video & text take all the width)

You can disable elements such as sidebars, set the content width etc.
